How effective is an over bored muzzle brake. Example 308 brake on 6.5 CM

Not sure you will notice any difference. It is more about the design of the brake than the bore.

They claim that 50 thou. and it starts to lose its effectiveness.
With the Big Magnums and Ultra Mags
I like to stay at 20 thou.
For others I can't tell the difference.
