When setting up my rifle profiles which altitude/mean sea kevel number should I use. Density altitude from my kestrel or actual altitude for my zero location?

Are you not using the gun profiles on the kestrel? What Leica product are you using?
In a hunting situation I have found the kestrel is too slow communicating with the bino. I would like to build the profile in the leica app to be as accurate as possible for reasonable hunting distances.
 
Assuming you are using a 100 yard zero. I would use the absolute pressure from the kestrel wherever you are hunting or shooting (not your zero location). When you put in the pressure and temp in the app it will give you a mean altitude.

I say this again assuming a 100 yard zero as your zero will change by maybe 0.02" at any elevation change but using a longer zero distance will likely change the results.
