I'd like to replace my factory Rem 700 300 RUM barrel with a new take-off
factory 375 RUM barrel. Do I really need to verify headspace ?
I know this might sound like a cavalier question, but, does the factory check
headspace on every new rifle that is assembled with production line parts ?

Yes you need to verify the head space on the new barrel. sometimes it works out but most often it does not fall within the needed .001 to .0025.

And yes they do have to check every rifle, but the head space may vary more than desired with any factory rifle no matter who makes it as long as it is safe (From .000 Up to .006 to .007 thousandths).
