About to cut my first chamber

I made a spider mounted at the back end of the spindle bore The muzzle end of the barrel is clamped and trued there with an indicator. The breech end is held with a 4 jaw chuck and indicated. I used a floating reamer holder, don't remember the brand. That setup worked for me. Take your time and use plenty of lubricant and should work well.
 
That's exactly what I had planned to do but my spindle bore is only 1" and when i looked all the green mountain blanks were bigger than that. I suppose I could turn between centers, but I have been planning to use an outboard spider.

You could always chamber between centers just to get some practice. I was glad I didn't do my first few chambers on a $400 barrel. Sure, they were ok but not perfect like I wanted. Looks like you are thinking it through
